We also utilize a bottom-up synthetic cell approach to study adhesion and polarization in a minimal cell-model. To this end we adhere Giant Unilamellar Lipid Vesicles (GUVs) onto micropatterned surfaces and look at the organization of cytoskeletal proteins. https://doi.org/10.1021/acssynbio.2c00516

When GUVs get deformed, the synthetic actin cytoskeleton can reorganize due to the outer cues. Using a minimal system to study this process allows to decouple interactions and behaviour of single cytoskeletal proteins, in ways not possible in living cells.
